旋毛虫肌幼虫培养细胞的扫描电镜观察
The ultrastructural observation of the cultured cells from Trichinella spiralis muscle larvae by scanning electron microscope
【摘要】 目的:观察旋毛虫(Trichinella spiralis,T1)肌幼虫培养细胞的超微结构。方法:分离旋毛虫肌幼虫细胞,进行体外传代培养,利用简单重复序列区间扩增(ISSR-PCR)标准引物对第3代培养细胞及肌幼虫基因组进行扩增,利用扫描电镜观察贴壁细胞的形态特征。结果和结论:旋毛虫肌幼虫第3代培养细胞和旋毛虫肌幼虫DNA的ISSR-PCR扩增产物均在1000bp和750bp处出现相同条带。扫描电镜下旋毛虫培养细胞的形态有多种类型,可分为圆颗粒形、多角形、三角扇形、蜂窝型、似分裂型及褶皱型等,其中以圆颗粒形细胞占多数。
【Abstract】 Aim:To observe the ultrastructure of the cultured cells from Trichinella spiralis(T.spiralis) muscle larvae.Methods:The cells from T.spiralis muscle larvae were isolated and cultured in vitro, the genome of the third passage cultured cells and muscle larvae were amplified by using ISSR-PCR. The morphological characters of the adhesive cells were observed by using scanning electron microscope. Results and Conclusion:The specific banding patterns (1 000 bp and 750 bp) from the cultured cells amplified by ISSR-PCR were the same as T.spiralis muscle larval DNA. Under scanning electron microscopy, the morphology of adhesive cells had several types consisting of round granular, polygonal, deltaic fan-shaped, alveolate, multiplication-like and drape cells, of which round granular cells were dominant.
