湖南芹菜中芹菜苷和3’-甲氧基芹菜苷含量的测定
Determination of Content of Apiin and 3’-methoxy Apiin in Hunan Celery
【摘要】 采用ODS色谱柱C18(150mm×4.6mm,5μm),以乙腈-水(23∶77)为流动相,在流速0.6mL/min,进样量10μL,柱温40℃条件下,PDA检测器在波长270nm测定湖南芹菜中芹菜苷和3’-甲氧基芹菜苷的含量。结果表明:不同地区不同部位芹菜中黄酮苷含量不同且差异较大,芹菜叶柄中黄酮苷含量明显低于芹菜叶中的含量。芹菜叶中芹菜苷、3’-甲氧基芹菜苷含量分别为0.12%~0.43%和0.06%~0.19%。芹菜叶柄中芹菜苷、3’-甲氧基芹菜苷含量分别为0.01%~0.04%和0.01%~0.03%。张家界实芹叶的芹菜苷和3’-甲氧基芹菜苷含量最高,分别为0.43%和0.19%,而张家界野芹菜中未检出芹菜苷和3’-甲氧基芹菜苷。
【Abstract】 The contents of Apiin and 3’-methoxy Apiin from Hunan Celery were determined by High Performance Liquid Chromatography (HPLC) systems equip with PDF detector and ODS C18 (150 mm ×4.6 mm, 5μm) chrom atographic analysis column. The column temperature was set at 40 °C and run on split mode with a constant flow of 0.6 mL/min (acetonitrile-water (23∶77). A sample in pouring volume was prepared by 10 μL. The detection wavelength was 270 nm. The results showed that the contents of flavone glycosides in different regions Celery varieties in Hunan were more difference. The contents of flavone glycosides in Celery leaves were higher than that in Celery stem. The contents of Apiin and 3’-methoxy Apiin in Celery leaves were 0.12% ~0.43% , 0.06% ~0.19% , respectively. The contents of Apiin and 3’-methoxy Apiin in Celery stem were 0.01%~0.04%, 0.01%~0.03%, respectively. The highest content of flavone glycosides from solid Celery leaves in Zhangjiajie was 0.43% (Apiin) and 0.19% (3’-methoxy Apiin) respectively. Apiin and 3’-methoxy Apiin from wild Zhangjiajie Celery were not found.
