应用G418催芽胁迫体系鉴定携带npt-Ⅱ基因的转育水稻纯系的研究
Study on Transgenic Homozygote of Rice with npt-Ⅱ Gene Identified by G418 System of Accelerating Germination Stress
【摘要】 研究了G418胁迫条件下转育后代的发芽特性和长根类比率,筛选获得了高抗G418的抗性株系,并进行其npt-Ⅱ基因及目的基因的检测与遗传分析。结果表明,转育株系如BM-3、BM-10、BM-13具有高发芽率(>90%)、高生根率(>95%)、低死亡率(<10%)和高长根比率(约为90%),经PCR结果发现这些株系内各单株都含有npt-Ⅱ基因及溶菌酶基因,确为转基因纯合体。进一步证实了G418胁迫催芽技术是可行和实用的,对于npt-Ⅱ基因转育纯系的筛选有重要意义。
【Abstract】 The germination characteristics and radication rate of transfer breeding progenies were studied under G418 stress.The strains resisting to G418 were bred,and the test and genetic analysis of npt-Ⅱgene and objective gene in it were conducted.The results showed that the transferring lines such as BM-3,BM-10 and BM-13 all had higher germination rate(more than 90%),high root-growing rate(more than 90%),low fatal rate(less than 10%) and high long length ratio(about 90%).The results of PCR indicated that these strains all were transgenic homozygote with npt-Ⅱgene and lysozyme gene,further confirmed the technique of accelerating germination under G418 stress is feasible and practical,and had significant to select the transgenic homozygote with npt-Ⅱgene.
