苦瓜花粉体外萌发培养基成分的优化
Optimization of composition of culture mediums for in vitro germination of bitter gourd pollen
【摘要】 应用二次正交旋转通用组合设计,研究了培养基中蔗糖、Ca2+和硼酸3浓度对苦瓜花粉体外萌发的影响,获得了相应的数学模型,解析了各因素对苦瓜花粉萌发率的主效应和互作效应,通过模型寻优获得了适于苦瓜花粉体外萌发的最优培养基配方.结果表明,3种成分对苦瓜花粉萌发率的影响从大到小依次为蔗糖、硼酸、Ca2+.在本试验条件下,苦瓜花粉萌发率大于73%的培养基配方为:蔗糖质量浓度8.0%~8.8%,硼酸浓度1.59~1.84mmol/L,Ca2+浓度2.98~3.44mmol/L.
【Abstract】 The effect of the concentration of sucrose, boric acid and Ca2+ in the cultural media on in vitro germination for bitter melon pollen was investigated, and a mathematic model was established involving the three factors and the germination rate by using the quadric orthogonal rotation general combination design. The major and interactive effect of the three factors on the germination rate were determined, and the optimum formulation of the cultural media for in vitro germination of bitter melon pollen was founded by analyzing the model. The results showed that the most important factor affecting the germination rate was sucrose, and then was boric acid, Ca2+ respectively. The optimum formulation of the cultural media for the germination surpassing 73% was:sucrose 8.0%~8.8%, boric acid 1.59~1.84 mmol/L and Ca2+ 2.98~3.44 mmol/L.
