山梨醇、ABA和干燥处理对玉米愈伤组织生长的影响
Effects of Sorbitol,ABA and Dehydration Treatment on the Growth of the Callus in Maize
【摘要】 以长期继代培养的玉米愈伤组织为材料,研究了山梨醇、ABA和干燥48h对愈伤组织生长的影响.结果表明,同一基因型随着继代次数的增多,愈伤组织的生长量呈下降趋势;除自交系87-1外,40g/L或50g/L质量浓度的山梨醇能够加快愈伤组织的生长;ABA处理对愈伤组织生长量的影响不明显,仅个别基因型的生长量比对照有所增加;48h干燥处理对所有基因型愈伤组织的生长都有明显的促进作用.比较山梨醇、ABA和干燥48h处理,干燥48h在提高玉米愈伤组织生长量方面效果最好,并且操作简便、没有基因型的差异,具有广泛的适用性.
【Abstract】 The effects of sorbitol,ABA and 48 h dehydration on the growth of long-term subcultured callus in Maize were investigated.The results are as follows:The growth capacity of the callus presented downtrend for the same genotype with the increase of subculturing times;Sorbitol of 40 g/L or 50 g/L concentration could accelerate the growth of the callus to all genotypes except inbred line 87-1;The effect of ABA on the growth capacity of the callus was not obvious except a few genotypes;48 h dehydration treatment could greatly promote the growth of the calli of all genotypes.Compared with sorbitol and ABA,the effect of 48 h dehydration treatment was the best in terms of increasing the growth capacity of the callus in maize,and this method could operate easily and there were no differences among various genotypes.So the 48 h dehydration treatment had extensive applicability.
