柳桉组织培养与工厂化育苗技术研究

【摘要】 以柳桉带腋芽的幼嫩茎段为外植体进行组培快繁,其启动培养、增殖培养、生根培养的最佳培养基依次为MS+6-BA2+NAA1、改良MS+6-BA0.2+NAA0.2+IBA0.8、改良MS+NAA0.2+IBA0.8,繁殖系数可达3以上,生根率可达85%以上;试管苗移栽前需炼苗10~15 d,移栽时间选在3、4、10、11月份移栽成活率最高,可达90%以上;柳桉萌发力强,试管苗移栽成活后长到一定高度经过采穗可促使腋芽不断萌发,生产上可利用此原理建立田间采穗圃,1株试管苗6个月内可供采穗10~12根;试管苗移栽前必须经过一个炼苗阶段,炼苗时间为10~15 d,移栽成活率可达90%以上;气温与柳桉扦穗的愈合、生根关系十分密切,在气温18~25℃时扦插,有利于插穗愈合生根;使用50 mg/L GGR溶液处理插穗,可使插穗生根提早5 d左右,同时可使生根率提高10%。

【Abstract】 The tissue culture and rapid propagation was test by explants of Eucalyptus saligna tender stem segments with axillary buds.The initiation culture was MS+6-BA2+NAA1,multiplication culture was MS+6-BA0.2+NAA0.2+IBA0.8 and rooting culture was MS+NAA0.2+IBA0.8.The propagation coefficient could reach more than 3 and rooting rate could reach more than 85%.The plantlets required hardening for 10 to 15 days before transplantation.The highest rate of transplant months were March,April,October and November,and could reach more than 90%.E.saligna has strong germination ability,the axillary buds of plantlets would geminated with cutting in some length of transplant.We could establish the cutting orchard with this principle of germination,and one plantlet could provide 10 to 12 cutting in six months.The plantlet must havea hardening-seedling period before transplantation,and the transplant rate of plantlet could reach more than 90% with hardening-seedling 10 to 15 days.The temperature was necessary for cutting healing and rooting.The suitable temperature for cutting healing and rooting was 18 to 25 ℃.Using 50 mg/L GGR could advance cutting roots five days and accelerate cutting rooting rate 10%.
