土霉素对半滑舌鳎肠道细菌耐药性影响与药物残留分析
Effects of intestinal microflora resistance following oral administration of oxytetracycline to Cynoglossus semilaevis Günther and residue analysis
【摘要】 研究半滑舌鳎(Cynoglossus semilaevis Gnther)肠道细菌耐药性与其肌肉中土霉素残留量之间的关系。养殖水温(22±1)℃,以125mg/(kg.d)剂量拌料投喂体重为(200±20)g的半滑舌鳎11d,利用土霉素选择培养基检测其肠道细菌耐药率的变化,同时检测其肌肉中土霉素残留量。结果表明,投药期间,在土霉素含量为16mg/L和160mg/L的选择性培养基上,肠道细菌耐药率迅速上升,第11天达到最大值,分别为(43.15±4.20)%和(18.64±0.45)%,远远高于对照组平均耐药率;土霉素在肌肉中的残留量为(2.23±0.40)~(3.14±0.15)mg/kg,呈上升趋势,投药期结束后残留量迅速下降,第11天降到检测限0.05μg/mL以下。休药期鱼体内的土霉素残留量随着肠道细菌的耐药率降低而迅速下降,并且形成了较好的拟合关系。试验对进一步利用鱼肠道细菌的耐药率来表征鱼体内土霉素的残留量提供了一种新的研究思路。
【Abstract】 To investigate the relationship between resistance rate of intestinal bacteria(RRI) and residual content of OTC in fish muscle(COM) in Cynoglossus semilaevis Günther,which were fed for 11 days with fish feedstuff to which the OTC was added with does of 125 mg/(kg·d) under the temperature of(22±1)℃.The RRIs and COM were investigated with the method of selective medium of OTC and HPLC,respectively.The results showed that in the selective mediums with OTC added,RRIs were sharply increased to the maximum of(43.15±4.20) % and(18.64±0.45) % with OTC concentration of 16 mg/L and 160 mg/L at the last day of adminstration,respectively.With the prolongation of time,RRIs were declined laggardly but still higher than the control level till 32 days after feeding deprivation;COM was increased to the maximum of(3.14±0.15) mg/kg after feeding for 7 days was decreased significantly after feeding deprivation,under the detection limit 10 days later.The results of the experiment indicated that there was no direct linear relationship between RRIs and COM in the administration period,COM reduced rapidly with the decrease of RRIs during the withdrawal period,and the fitting curve equation was obtained.So the RRIs supplied a new way of thinking to represent the COM.
