高血压性大鼠脑出血血肿量对NF-κB表达的影响
Effect of the volume of hematoma of hypertensive intracerebral hemorrhage in rats on expression of NF-κB
【摘要】 目的观察高血压性大鼠脑出血后血肿周围组织核因子-κB(necular factor kappa B,NF-κB)的表达,分析其表达与血肿量及出血时间的关系。方法成年SD大鼠100只,体重250~300g。随机分为对照组、脑出血A组、脑出血B组。大鼠高血压性脑出血模型采用双侧肾动脉前支部分热凝,立体定位仪下自体血脑内注入法建立。3组在相应时间点用免疫组化方法观察NF-κB在细胞内的表达情况。结果大鼠高血压性脑出血后1h后NF-κB即开始表达,24h达到高峰,持续至72h开始下降,并且血肿量大的脑出血组(脑出血B组)相应时间点NF-κB的表达较血肿量小的脑出血组(脑出血A组)明显增高(P<0.05)。结论高血压性脑出血后早期即有NF-κB的表达,并且随血肿量的增加表达也增加,从而加重了脑出血后周围脑组织的继发性损伤。
【Abstract】 Objective To observe the effect of the volume of hematoma of hypertensive intracerebral hemorrhage in rats on expression of nuclear factor-κB (NF-κB),and to investigate the relationship between the expression NF-κB and volume of hematoma as well as bleeding time. Methods 100 adult SD rats,weighted 250~300 g,were randomly divided into three groups:control group,cerebral hemorrhage group A and cerebral hemorrhage group B. The hypertensive intracerebral hemorrhage (HICH) models of rats were induced by the injection of auologous blood with stereotactic techniques after partial coagulation of the anterior branches of bilateral renal arteries. The expression of NF-κB was detected by immunohistochemical method. Results The expression of NF-κB could be found 1 hour after HICH in rats,and which reached peak at 24 hours and lasted to 72 hours,then began to decrease.Moreover,the expression of NF-κB in cerebral hemorrhage group B at corresponding time points was significantly higher than those in cerebral hemorrhage group B. Conclusion The expression of NF-κB can be found at early period after HICH,which increases with the increase of volume of hematoma so as to aggravate secondary injury of pericerebral tissues.
