侧脑室注射钩藤碱增加大鼠心血管相关核团中c-fos的表达

【摘要】 目的研究侧脑室注射钩藤碱(Rhy)对大鼠脑内心血管相关核团中c-fos表达的影响。方法应用Fos蛋白免疫组化技术。结果侧脑室注射Rhy诱发脑干的延髓头端腹外侧核(RVL)、蓝斑核(LC)、旁巨细胞外侧核(iPGL),下丘脑的室旁核(PVN)、视上核(SON)等多个部位的心血管中枢出现大量Fos样免疫反应(FLI)神经元。L型-钙通道开放剂Bayk 8644可明显减弱Rhy的效应。蛋白酪氨酸磷酸酶抑制剂正矾酸钠也可明显减弱Rhy的效应。结论Rhy可兴奋脑内多个心血管相关核团的神经元,其机制可能与阻断神经元上的钙通道并进而抑制蛋白质磷酸化有关。

【Abstract】 Objective To observe the effects of intracerebroventricular(ICV) injection with rhynchophylline(Rhy) on the expression of c-fos gene in rat brain nuclei involved in cardiovascular regulation.Methods Immunohistochemistry was used to detect the expression of c-fos.Results Following ICV injection with Rhy(50 nmol/kg),Fos-like immunoreactivity(Fos-LI) neurons were obviously increased in various brain areas of the rats,including the nucleus of the rostralventrolateral reticular nucleus,locus coeruleus,parabrachial nucleus and nucleus paragigantocelluaris laterialis in brainstem,paraventricular nucleus,and supraoptic nucleus in hypothalamus as well as the lateral habenular nucleus in the forebrain.Pretreatment with Bay K8644-an activator of L-type Ca2+ channel,could significantly reduce the effects of Rhy(50 nmol/kg) on the induction of Fos-LI in brain.Pretreatment with Na3VO4——a specific protein tyrosine phosphatase(PTPase) inhibitor,could reduce the effects of Rhy too.Conclusion The intracerebroventricular injection with Rhy can activate the neurons in the forebrain,hypothalamus and brainstem,and the effects may be mediated by a decrease in Ca2+ influx,in which protein tyrosine kinase(PTK) is involved.
