成骨细胞移植对实验性家兔股骨头坏死的治疗作用
THERAPEUTIC EFFECTS OF OSTEOBLASTS TRANSPLANTATION ON EXPERIMENTAL NECROSIS OF FEMORAL HEAD IN RABBITS
【摘要】 目的比较骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)及其成骨诱导后移植,对实验性家兔股骨头坏死的治疗作用。方法采用24只成年家兔,随机分为BMSCs移植组(A组)与BMSCs诱导分化的成骨细胞移植组(B组),将细胞移植于坏死股骨头的钻孔区内。对术后2、4、6、8周的股骨头标本,进行X线、组织学和骨形态发生蛋白(bone morphgenetic protein,BMP)免疫组织化学观察和图像分析。结果①X线观察显示,随着时间的进展,A、B2组钻孔区密度逐渐增高,6周时出现骨小梁结构,8周呈现正常的骨小梁结构。②组织学观察表明,在A、B 2组钻孔区内,术后2周时有大量的成骨细胞;4周时充满骨小梁;8周时骨小梁趋于成熟,2组间骨小梁面积的图像分析,其差异无统计学意义(P>0.05)。③免疫组织化学观察显示,在A、B2组术后2周时,移植的BMSCs呈现BMP免疫阳性反应;然而,向成骨细胞诱导分化的BMSCs移植后,只有部分细胞出现BMP免疫阳性反应。结论 BMSCs及其成骨诱导后移植,对实验性家兔股骨头坏死的治疗,呈现相似的结果;股骨头内部的微环境,对BMSCs具有明显的向成骨细胞诱导分化作用。
【Abstract】 Objective To compare the repair effect of bone marrow mesenchymal stem cells (BMSCs) and BMSCs after osteogenic induction transplantation on experimental necrosis of femoral head(ENFH) in rabbits. Methods Twenty four adult rabbits were randomly divided into BMSCs group(A) and osteoblast group(B). The cells were transplanted into the drilled hole area (DHA) of femoral head. The femoral head specimens were observed with radioscopy and microscope for histology, bone morphgenetic protein (BMP) immunohistochemistry and image analyzing respectively at 2,4,6,8 weeks after operation. Results ①Radioscopic observation: In the two groups,the density in the DHA increased at 4 weeks. Trabecula appeared in the DHA at 6weeks. At 8 weeks, more normal trabecula formed in the DHA. ②microscopic observation: In the two groups, there were a large amount of BMSCs in the DHA at 2 weeks. And then DHA were filled of trabecula at 4 weeks. At 8 weeks,the trabeculae matured and bone marrow appeared in the intertrabecular space. There was no statistical difference between group A and group B in trabeculae area. ③Immunohistochemical observation: The implanted BMSCs in all area were positive for BMP, Induced BMSCs in partial area were positive for BMP immune response at 2 weeks after operation. Conclusion There was resemble repair effect in treating ENFH in rabbits with BMSCs and induced BMSCs transplantation. As to osteogenic induction BMSCs,the inducing effect in micro-environment of femoral head was more effective than induction in vitro.
