【摘要】 目的观察基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)、组织基质金属蛋白抑制剂-2(tissue inhibitor of metalloproteinase-2,TIMP-2)在血管瘤不同分期中的表达。方法取承德医学院附属医院1998年1月—2005年12月期间血管瘤手术切除的标本共60例及正常带血管皮肤手术切除标本10例,患者家属知情同意。①实验分组,根据 Mulliken 标准进行病理诊断并分类,所有标本共分4组,增生组22例,退化组20例,退化完成组18例,正常带血管皮肤组织作为对照组10例。②采用免疫组织化学 SP 法对各组标本的 MMP-2、TIMP-2进行染色。③实验评估,以正常血管上皮细胞和肿瘤细胞胞浆出现棕黄色颗粒为 MMP-2、TIMP-2的阳性表达。结果随着血管瘤病理时期的变化,MMP-2、TIMP-2出现明显不同的表达。增生组 MMP-2的阳性表达率明显高于其他各组(P<0.01),TIMP-2的阳性表达率低于其他各组(P<0.01),随着血管瘤的退化 MMP-2的阳性表达率逐渐下降,TIMP-2的阳性表达率逐渐提高,至退化完成期时二者与对照组无明显差别。MMP-2与 TIMP-2的表达呈负相关。结论 MMP-2、TIMP-2在血管瘤分期中起重要作用,其表达水平与血管瘤的病理分期有密切关系。更多还原

【Abstract】 Objective To observe the expressions of matrix metalloproteinase-2 (MMP-2), tissue inhibitor of metalloproteinase-2 ( TIMP-2 ) in different period of hemangioma. Methods Sixty specimens of hemangioma tissue excised in operation and 10 normal skin with blood vessels removed in operation were harvested from affiliated hospital of chengde medical college between January 1998 and December 2005 after the informed consent was obtained from the patients.① The pathologic diagnosis was made according to Mulliken standard under microscope,all the specimens were divided into 4 groups,which was proliferated period group ( n =22),degenerated period group( n 20),completed degeneration period group (n = 18),and 10 normal skin with blood vessels were taken as control group.②The MMP-2,TIMP-2 were stained by SP method.③ The expression of the MMP-2,TIMP-2 in each group was detected with immunohistochemistry.The positive expression was that the brown and yellow granule appearance in the endochylema of the normal Haemal- epithelial cell and the tumor cell. Results The expression of the MMP-2,TIMP-2 changed with hemangiomatous pathologic period. The positive expression of the MMP-2 in proliferated period was significantly higher than that in other groups,and the positive expression of the TIMP-2 was lower than that in other groups;with the degeneration,the positive expression of the MMP-2 was gradually decreased,the positive expression of the TIMP-2 is opposite.When hemangioma completely degenerated,the expression of the two indexes was nearly the same as the control group.The expression of the MMP-2 was negatively related with that of TIMP-2.Conclusion The MMP-2,TIMP-2 play very important roles in different period of hemangioma,and the levels of their expression are related with hemangiomatous period.更多还原
