肝癌组织中Survivin和NF-κB的表达及其与细胞增殖和凋亡的关系
EFFECTS OF Survivin AND NF-κB EXPRESSION ON APOPTOSIS AND PROLIFERATION IN HEPATOCELLULAR CARCINOMA
【摘要】 目的分析 Survivin、核因子-κB(nuclear factor-kappaB,NF-κB)在肝组织中的表达及其与肝癌细胞增殖和凋亡间的相互关系。方法应用免疫组织化学链霉卵白素-生物素-辣根过氧化物酶复合物法(streptavidinperoxidase conjugated method,SP)检测44例肝细胞癌(hepatocellular carcinoma,HCC)组织、25例癌旁相应组织、9例正常肝组织中 Survivin、NF-κB、Ki-67蛋白的表达情况,用末端脱氧核苷酸转移酶介导的 dUTP 缺口标记(terminal deoxynueleotidyl trans[erase-mediated dUTP nick end labeling,TUNEL)法检测凋亡细胞,并计算增殖指数(proliferation index,PI)和凋亡指数(apoptosis index,AI)。结果 HCC 组织中 Survivin、NF-κB蛋自阳性表达率显著高于癌旁组织和正常肝组织(P<0.01);HCC 组织中 Survivin 阳性表达病例平均 PI 值(35.19±16.53)%显著高于阴性表达者(16.38±10.11)%(P<0.05),Survivin 阳性表达病例平均 AI 值(3.15±1.52)%显著低于阴性表达者(5.24±2.27)%(P<0.05);HCC 标本中 NF-κB阳性表达病例平均 PI 值(29.88±13.71)%与阴性表达者(24.15±10.87)%比较无统计学意义(P>0.05),NF-κB阳性表达病例平均 AI 值(2.89±1.42)%显著低于阴性表达者(4.91±2.04)%(P<0.05);NF-κB与 Survivin 的表达呈显著正相关(P<0.05)。结论 Survivin 和 NFκB在 HCC 中高表达。Survivin 通过促进细胞增殖、抑制细胞凋亡的方式共同影响肝癌的生物学特性。而 NF-κB通过抑制细胞凋亡的方式影响肝癌的生物学特性,对肝癌的细胞增殖无显著影响。NF-κB与 Survivin 的表达呈正相关。
【Abstract】 Objective To analyze the relationship between Survivin or nuclear factorkappaB (NF-κB) protein expression and hepatocellular carcinoma cells proliferation or apoptosis. Methods The expressions of Survivin,NF-κB and Ki-67 protein were tested using immunohistochemistry streptavidin peroidase conjugated method (SP) method,and the cell apoptosis was tested by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) method in hepatocellular carcinoma (HCC) tissues of 44 patients,para-cancerous tissues of 25 patients with HCC and 9 with normal liver.Subsequent proliferation index(PI) and apoptosis index(AI) were calculated.Results The positive rate of Survivin and NF-κB protein expression was higher than para-cancerous tissues and normal liver tissues (P<0.05);The average of PI in cases of survivin protein positive (35.19±16.53)% was higher than the negative (16.38±10.11)% in HCC (P<0.05).The average of AI in cases of Survivin protein positive (3.15±1.52)% was lower than negative (5.24±2.27)% (P<0.05);The average of PI in NF-κB protein positive cases was (29.88±13.71)%,and (24.15±10.87)% in negative ones in HCC. There was no significant difference between the two groups (P>0.05).In NF-κB protein positive cases,the average of AI (2.89 ± 1.42)% was lower than (4.91± 2.04)% in negative ones (P<0.05);Survivin protein expression was positively correlated with NF-κB (P<0.05). Conclusion There was high level expression of Survivin and NF-κB protein in HCC tissues.The expression of survivin might influence the biological property by both inhibiting cell apoptosis and promoting.proliferation.NF-κB played an important role in the cell apoptosis and did not significantly influence the cell proliferation of HCC.The expression of Survivin protein was positively correlated with NF-κB.
