【摘要】 目的探讨PPARγ和MMP-7在食管鳞状细胞癌发生、发展中的作用及生物学意义。方法采用免疫组织化学SP法检测PPARγ和MMP-7蛋白在40例食管鳞状细胞癌、25例非典型性增生组织和13例正常黏膜中的表达。结果PPARγ在癌组织中的阳性表达率为52.5%,高于不典型增生组织(20%)和正常黏膜(7.7%)(P<0.05);MMP-7在癌组织中的阳性表达率为62.5%,高于不典型增生组织(28.0%)和正常黏膜(0%)(P<0.05)。PPARγ的表达与细胞的分化程度、淋巴结转移相关,MMP-7的表达与浸润深度和淋巴结转移相关。PPARγ和MMP-7的表达呈正相关。结论PPARγ和MMP-7基因在食管鳞癌中具有高表达,且表达具有肿瘤特异性,提示PPARγ和MMP-7在食管鳞癌的生长和侵袭转移过程中有非常重要的作用。更多还原

【Abstract】 Objective To investigate the expression and biological significance of peroxisome proliferators-activated receptor γ(PPARγ) and matrix metalloproteinase-7(MMP-7) in esophageal carcinoma. Methods The expression of PPARγ and MMP-7 was examined by SP immunohistochemical stain(IHC) in 40 cases of esophageal carcinoma specimens,25 cases of dysplasia and 13 cases of normal tissues. Results The positive ratio of PPARγ in esophageal carcinoma (52.5%) was significantly higher than that in dysplasia(20%) and normal tissues (7.7%) (P<0.05).The positive ratio of MMP-7 in esophageal carcinoma (62.5%) was significantly higher than that in dysplasia(28.0%) and normal tissues (0%) (P<0.05).High level of PPARγ was significantly correlated with poor differentiation and lymph node metastasis(P<0.05).High level of MMP-7 was significantly correlated with lymph node metastasis and fibromembranous infiltration (P<0.05).There was a positive correlation between MMP-7 and PPARγ. Conclusion The high expressions of PPARγ and MMP-7 could be involved in oncogenesis and metastasis of esophageal squamous cell carcinoma.更多还原
