【摘要】 目的:研究细胞周期素D1(Cyclin D1)基因多态性与胃癌易感性的关系,同时探讨Cyclin D1基因多态性及其与幽门螺杆菌(Hp)感染联合作用对胃癌发病风险的影响。方法:采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法检测115例胃癌与112例慢性胃炎患者(对照组)Cyclin D1基因第870位核苷酸A/G(A870G)多态性,应用ELISA方法检测上述人群中Hp感染率。结果:胃癌组Cyclin D1基因型频率与对照组比较,差异有显著性(P<0.05)。与AG及GG基因型携带者之和相比,AA基因型携带者患胃癌的相对风险度(OR)为2.60,95%可信区间(CI)为1.47-4.58。Hp阳性胃癌组Cyclin D1基因型频率与Hp阳性对照组比较,差异亦有显著性(P<0.05)。与AG及GG基因型携带者之和相比,携带AA基因型的Hp阳性者患胃癌的OR为3.82,95%CI为1.92-7.61。结论:Cyclin D1基因多态性与胃癌遗传易感性有关,携带Cyclin D1 AA基因型个体患胃癌的危险性增高。Hp感染和Cyclin D1基因多态性有协同促进胃癌发生的作用。更多还原

【Abstract】 Objective: To investigate the relationship between Cyclin D1 gene polymorphism and susceptibility of gastric cancer,and to evaluated the combined effect of Cyclin D1 gene polymorphism and Helicobacter pylori(Hp) infection on gastric cancer.Methods: By polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP),the Cyclin D1(A870G) genotyping was performed among 115 cases with gastric cancer and 112 controls with chronic gastritis.ELISA was adapted to examine Hp infection of the above patients.Results: There was a significant difference of Cyclin D1 genotypes frequencies between patients with gastric cancer and controls(P<0.05).The subjects carrying AA genotype were at an increased risk of gastric cancer compared to the subjects carrying A/G and GG genotype,and the odds ratio(OR) was 2.60(95%CI,1.47-4.58).In patients with Hp infection,the difference of Cyclin D1 genotypes frequencies between patients with gastric cancer and controls was statistically significance(P<0.05).The risk for gastric cancer was higher in AA genotype carriers with Hp infection than in A/G and GG genotype carriers with Hp infection,with an OR of 3.82(95%CI,1.92-7.61).Conclusion: The Cyclin D1 gene polymorphism is related to the genetic susceptibility of gastric cancer.Individuals with Cyclin D1 AA genotype show an increased risk for gastric cancer.Association of the Cyclin D1 AA genotype with Hp infection could significantly increase the gastric cancer risk.更多还原
