东方百合组培快繁技术研究
Study on Tissue Culture Technique of Eastern Lilium Brownii for Rapid Propagation
【摘要】 用东方百合的鳞片、叶片、茎段、根尖为外植体,系统研究不同培养基、不同质量浓度的植物激素在东方百合组织培养快速繁殖中的相互作用,筛选出最佳诱导和增殖的培养因素组合.同时进行试管种球、试管苗无土栽培技术的研究.研究表明东方百合鳞球茎的鳞片为组培快繁较好的外植体,MS+NAA(0.2mg/L)+6-BA(2.0mg/L)为根诱导最佳培养基,MS+NAA(0.5mg/L)+6-BA(1.5mg/L)为最佳培养基,MS/2+NAA0.3~0.5(mg/L)为最佳生根培养基.进口伯爵泥岩525#∶珍珠岩∶蛭石=5∶1∶1混合是试管苗炼苗最人士栽培基质.
【Abstract】 Scales,leaves,stem segments and root tips of Eastern Lilium Brownii were used as ex-plant in the study.Mutual effects of different culture medium,growth regulators and/or different concentration were systematically studied and the most favorable media combination of culture factors for callus inducing and subculture for rapid propagation were selected in relevant experiment.Meanwhile,soilless culture technique research of plantlet and seed ball produced in vitro was also carried out.The experiment showed that: the best ex-plant was scales;the most favorable root inducing medium formula was MS+NAA(0.2 mg/L)+6-BA(2.0 mg/L),1/2MS+NAA0.3~0.5 for root growth proliferation.The best soilless culture medium for plantlet produced in vitro was the mixture of perlite,vermiculate peat with a proportion of 5∶1∶1.
