RP-HPLC法测定麻黄附子细辛汤水煎液中麻黄碱的含量

【摘要】 目的建立测定麻黄附子细辛汤水煎液中麻黄碱含量(以盐酸麻黄碱计)的反相高效液相色谱法。方法色谱柱:Diamonsil-C18柱(200mm×4.6mm);流动相:乙腈:水(含0.02mol.μL-1磷酸二氢钾和0.2%三乙胺)(10:90);流速:1.0mL.min-1;检测波长:210nm;柱温:27℃。结果样品中盐酸麻黄碱的色谱峰与其他组分完全达到基线分离,回归方程为:y=2229.4854x-22.2885,r=0.9998(n=6),线性范围0.1776-1.3320μg,平均加样回收率为101.45%(RSD=1.9%,n=6)。结论所建立的测定方法快速简便,重现性良好,可用于麻黄附子细辛汤水提物中麻黄碱的含量测定。

【Abstract】 OBJECTIVE To establish a RP-HPLC method for the determination of ephedrine(determined as ephedrine hydrochloride)in the water decoction of mahuang-fuzi-xixin-soup.METHODS The sample was separated on a Diamonsil-C18(200mm×4.6mm)with the mobile phase consisting of acetonitrile and water(containing 0.02mol·L-1 potassium dihydrogen phosphate and 0.2% triethy lamine)(10:90) at a flow rate of 1.0mL·min-1.The detection wavelength was 210nm,and the column was kept at 27℃.RESULTS Chromatographic separation of ephedrine hydrochloride and the co-existed compounds achieved baseline resolution,y=2229.4854x-22.2885,r=0.9998(n=6),the calibration was linear in the range of 0.1776-1.3320μg,and the average recovery of ephedrine hydrochloride was 101.45%(RSD=1.9%,n=6).CONCLUSION The method is rapid,simple and reproducible and can be used for the quantity detection of ephedrine in the water decoction of Mahuang-Fuzi-Xixin-soup.
