【摘要】 目的:研究体外缺氧对HT-29人结肠癌细胞鞘氨醇激酶-1(SK-1)表达及细胞迁移的影响。方法:模拟体外缺氧环境,应用RT-PCR及Western blot检测缺氧对HT-29细胞SK-1表达的影响,以及观察细胞迁移能力的情况。结果:缺氧状态下HT-29细胞SK-1 mRNA及蛋白表达增强,细胞迁移能力增强;且U0126具有抑制SK-1表达增强的作用。结论:缺氧状态下HT-29细胞迁移能力和SK-1基因表达存在一定关系。

【Abstract】 Objective: To study the effect of hypoxia in human colon carcinoma HT-29 cells(HCCHC) on sphingosine kinase-1(SK-1) expression and cell migration in vitro.Methods: RT-PCR and western blot were used to detect SK-1 expression in HCCHCs under artificially simulated oxygen deficient environment.Flourescence staining was adopted to observe the cell migration.Results: Under the oxygen deficient condition,expression of SK-1 mRNA and protein increased,so was the cell migration.U0126 showed an inhibitory effect on the expression increase.Conclusion: The migration of human colon carcinoma HT-29 cells was related to the mRNA level of SK-1 under the hypoxic condition.
