沙田柚花粉管S1-RNase免疫胶体金定位研究

【摘要】 以10年生沙田柚结果树为材料,应用免疫胶体金标记抗体技术检测沙田柚(Citrus grandis var.Shatinyu Hort)自花授粉后3d、1/2花柱中花粉管S1-RNase免疫胶体金颗粒的定位分布。结果表明,在自交花粉管的内质网、细胞质和细胞纤维壁上有胶体金颗粒分布,而对照的内质网、细胞质和细胞纤维壁上均未发现有胶体金颗粒分布。因此,自交花粉管中S1-RNase分别定位于内质网、细胞质和细胞纤维壁上,该结果为沙田柚自交不亲和机理研究提供了参考依据。

【Abstract】 Fruiting tree of Cirus grandis var.Shatinyu Hort planted for 10 years was used as material in the experiment,and immunogold(GRA-Au complexes) labeled antibody techniques were used to identify the localization of S1-RNase in pollen tube of 1 / 2 style site of Cirus grandis var.Shatinyu Hort three days after self-pollination.The results showed that immunogold particles were distributed in endoplasmic reticulum,cytoplasm and cell fiber wall in proper order,respectively,while there was no immunogold particle distributed in those of the control.Therefore,S1-RNase of pollen tube was dis-tributed in endoplasmic reticulum,cytoplasm and cell fiber wall,which would provide references for studying the mechanism of self-incompatibility of Cirus grandis var.Shatinyu Hort.
