石墨炉原子吸收法检测胎盘镉水平

【摘要】 建立了硝酸消化、石墨炉原子吸收法(GFAAS)测定胎盘中镉含量的方法。取0.3 g胎盘于110℃烤箱烘烤1 h,在80℃水浴下HNO3、H2O2顺序消化1 h,消化后定容至2 mL;D2灯校正背景,样本基体匹配、标准曲线法测定。结果表明,此法相对标准偏差<10%,回收率为89.4%~91.2%,此检测方法稳定,简便可行,胎盘取材方便,对新生儿无损伤,对新生儿镉负荷的检测评估具有一定的意义。

【Abstract】 To establish a method for determination of cadmium in placenta of newborns through nitric acid digestion by graphite furnace atomic absorption spectrometry(GFAAS),fresh placentas were collected and stored at-20℃.After being baked for at least 1 h at 110℃,0.3 g placenta was digested by nitric acid and hydrogen peroxide in turn at 80℃ under water-bath condition for 1 h and then metered the whole volume to 2 mL.After correcting the background with D2 lamp,specimen basal corpuscle matched standard curve was used to detect cadmium content.The relative standard deviation was less than 10%.Recovery rate was between 89.4%~91.2%.This method was stable and efficient.In addition,it is convenient to make use of placenta.There is no impairment in neonates.There is important significance using this method to evaluate chromium load in neonates.
