Smac/DIABLO在大鼠缺血皮质的变化及意义
Characterization of Smac/DIABLO Expression in Rat brain after Ischemic Brain Injury
【摘要】 目的:观察大鼠缺血区皮质Smac/DIABLO的动态变化,探讨Smac/DIABLO在缺血后神经元凋亡过程中的可能作用机制。方法:SD大鼠130只,随机分为对照组50只和模型组80只,采用RT-PCR、免疫组化和Western blot方法分别从mRNA和蛋白水平测定2组大鼠缺血区皮质在脑缺血后不同时间点Smac/DIABLO的表达变化。结果:Smac/DIABLO基因转录水平和蛋白表达的改变呈时间依赖性。RT-PCR显示模型组大鼠在脑缺血后3 h就有明显的Smac/DIABLO mRNA表达上调,持续至24 h后逐渐下降,48 h时恢复至缺血前水平;免疫组化和Western blot显示在脑缺血后3 h Smac/DIABLO蛋白表达增加,6 h蛋白表达至高峰(P<0.01),48 h基本恢复至缺血前水平。Smac/DIABLO mRNA转录水平改变提前于Smac/DIABLO蛋白表达。结论:Smac/DIABLO可能在脑缺血后神经细胞凋亡过程中起重要作用。
【Abstract】 Objective: To observe the temporal expression of Smac/DIABLO in rat brain after ischemic brain injury and to explore its potential role in mediating neuronal apoptosis.Methods: The expression of Smac/DIABLO in 80 rats after ischemic brain injury was investigated by RT-PCR,immunohistochemistry and western blot at hours 3,6,12,24,48 after ischemia.Results: The expression of Smac/DIABLO showed a time-dependent change after cerebral ischemia.The expression of Smac/DIABLO mRNA increased at 3 h after cerebral ischemia which persisted until 24 h and recoverd to normal level at 48 h.The protein expression revealed the similar change except that the increase peaked at 6 h after ischemia.Concluision: Smac/DIABLO may be involved in the process of neuronal apoptosis following ischemic brain injury.
