犬自发性前列腺增生组织中雄激素和5α-还原酶活性研究
Study on the Androgen Level and 5α-reductase Activity in Prostate Tissues of Dogs With Benign Prostatic Hyperplasia
【摘要】 目的:在哺乳类动物中,只有人类老年男性和老年雄性犬可自发发生良性前列腺增生(BPH),两者的发病机制相似。在对老年雄性犬BPH动物模型病理学和血生化分析基础上,研究犬BPH组织中雄激素(睾酮、双氢睾酮)水平、5α-还原酶活性。方法:选择6~13岁杂种老年雄性家犬,经肛门指检、直肠探头超声和手术直观下测量前列腺体积,后经病理确认7只BPH和5只无BPH老年雄性犬为实验对象。前列腺组织制备匀浆,放射免疫法测定睾酮和双氢睾酮水平,酶化学分析法测定5α-还原酶2个同功酶活性。结果:与无BPH犬相比,BPH犬血中睾酮及雌二醇(E2)水平差异无统计学意义(P>0.05);前列腺组织中睾酮水平差异也无统计学意义(P>0.05),但双氢睾酮水平升高(P<0.05),5α-还原酶II型同功酶活性显著升高(P<0.01)。结论:自发性BPH犬前列腺组织中5α-还原酶活性升高,双氢睾酮积蓄,是人类BPH研究有用的实验动物模型。
【Abstract】 Objective:Benign prostatic hyperplasia (BPH) can develop spontaneously only in older men and aged male dogs among mammals,and the pathological mechanisms of them are similar. This study is to investigate the androgen (testosterone and dihydrotestosterone) levels and 5α-reduetase activities based on the pathological and blood biochemical analysis of aged male dogs with BPH. Methods:Hybrid aged male dogs (6-13 yeas old) were examined by digital rectal examination,transrectal ultrasound and volume measurement of prostate in surgery. 7 dogs with BPH were finally confirmed by pathological examination,and 5 normal aged male dogs were recruited as control in this study. Prostate tissue homogenate was prepared. Testosterone (T) and dihydrotestosterone (DHT) levels were determined by radioimmunoassay (RIA),and isoenzyme activities of 5α-reductease were measured using zymochemistry assay. Results:Compared with the control dogs,the differences of sera levels of T and estradiol (E2) in dogs with BPH were not statistically significant (P>0.05),and there were no significant differences of T levels in prostate tissue (P>0.05),while DHT levels increased obviously (P<0.05),and the activities of type II isoenzyme of 5α-reductase elevated significantly (P<0.01). Conclusion:Aged male dogs with spontaneous BPH are useful experimental animal models of BPH research for the raised activities of 5α-reductase and the accumulation of DHT in prostate tissue.
