高亲和力抗膀胱癌单链抗体筛选及亲和力测定

【摘要】 以膀胱癌细胞系BIU-87膜抗原为亲和底物,从抗膀胱癌噬菌体人源化单链抗体库中筛选出4个高亲和力改形单链抗体.ELISA结果和NH4SCN洗脱法数据显示,它们对膀胱癌细胞膜抗原的亲和活性显著高于鼠源单链抗体.同时以多种蛋白检验了其亲和作用的特异性.

【Abstract】 Through 3 rounds of panning with BIU-87 cell associated membrane antigen,we screened the phagemid library for high affinity humanized scFv and 4 clones were obtained as a result.Data from measurement of thiocyanate elution ELISA showed that they had more binding-activity to BIU-87 cell associated membrane antigen than that of starting mouse scFv.And then,we tested and verified their special affinity by sorts of other proteins.
