果梅nSSR、cpSSR引物在李属果树上的通用性分析

【摘要】 用来自果梅的17对nSSR引物和13对cpSSR引物,对李属果树桃、李、梅、杏、樱桃5个树种的24个基因型进行了扩增,以分析其在李属不同果树上的通用性。结果显示,有13对nSSR引物和13对cpSSR引物得到多态性扩增,分别占所用引物的76.5%和100%,平均PIC值分别为0.591和0.654。其中,9对nSSR引物和11对cpSSR引物具有高多态性信息含量,可用于李属果树基因组学和遗传育种方面的研究,显示出果梅SSR引物在李属类果树中具有很好的通用性。

【Abstract】 Seventeen nSSR and thirteen cpSSR primer pairs developed from Japanese apricot(Prunus mume Sieb.et Zucc.) were employed to amplify microsatellites in 24 genotypes of peach,plum,Japanese apricot,apricot and cherry for detecting the universality of these primers used in Prunus fruit trees.The results were as follows:polymorphic DNA bands were amplified by PCR with 13 nSSR and 13 cpSSR primer pairs which occupied 76.5 percent and 100 percent among the total screened ones.The average polymorphism information contents(PIC) of nSSR and cpSSR markers were 0.591 and 0.654 respectively.High polymorphism was displayed with 9 nSSR and 11 cpSSR primer pairs that could be applicable to study genome,genetics and crop breeding of Prunus fruit trees.This study demonstrated that 9 nSSR and 11 cpSSR primer pairs developed from Japanese apricot had universality in detecting DNA polymorphism in Prunus fruit trees.
