c-Met与Fas在胃癌组织中的表达及其相关性
EXPRESSIONS OF c-Met AND Fas PROTEINS IN GASTRIC CARCINOMA AND THEIR RELATIONSHIP
【摘要】 目的探讨c-Met和Fas在胃癌中的表达、临床意义及其相关性。方法采用免疫组化方法检测c-Met和Fas在正常胃黏膜(n=17),胃癌旁组织(n=51)及胃癌组织(n=51)中的表达。结果在正常胃黏膜、胃癌旁组织及胃癌组织中,c-Met阳性率呈递增趋势,组间差异有统计学意义(χ2=19.881,P<0.01);Fas阳性率有递减趋势,组间差异有统计学意义(χ2=9.590,P<0.01)。χ2检验显示c-Met在浸润深度和淋巴结转移组内阳性表达率组内差异有统计学意义(P<0.05),而在年龄、性别和组织分型组内差异无统计学意义(P>0.05);Fas在浸润深度组内阳性表达率组内差异有统计学意义(P<0.05),而在年龄、性别、组织分型和淋巴结转移组内差异无统计学意义(P>0.05)。Spearman等级相关分析显示c-Met和Fas表达呈正相关(rs=0.599,P<0.01)。结论c-Met过表达和Fas表达下调与胃癌的发生发展及浸润转移有关,c-Met表达可能是胃癌细胞逃逸Fas介导的细胞凋亡的一种机制。
【Abstract】 Objective To investigate the expression and clinical significance of c-Met and Fas gene in human gastric carcinoma and their relationship.MethodsThe expressions of c-Met and Fas were detected by immunohistochemistry among normal gastric mucosa(n=17),adjacent normal tissue(n=51) and gastric carcinoma(n=51).ResultsThe positive expression rate of c-Met protein among normal gastric mucosa,adjacent normal tissue and gastric carcinoma was gradually increased,and statistical differences among the three groups were significant(χ2=19.881,P<0.01);the positive expression rate of Fas protein was gradually decreased with significant statistical difference(χ2=9.590,P<0.01).The expression of c-Met protein was related to the malignant progression of gastric carcinoma,depth of invasion and lymph node metastasis(P<0.05),but not correlated with the patient’s age,gender and differential grade(P>0.05).The expression of Fas was related to depth of invasion(P<0.05),but not correlated with the patient’s,age,gender,differential grade and lymph node metastasis(P>0.05).In addition,the expression of c-Met protein was positively correlated with that of Fas protein(rs=0.599,P<0.01).ConclusionThe overexpression of c-Met protein with Fas downregulation plays an important role in the development of gastric carcinoma,and c-Met expression may be a possible mechanism for gastric carcinoma cells to escape the Fas-mediated apoptosis.
