光照对姜黄素诱导人乳腺癌MCF-7细胞凋亡的影响

【摘要】 目的研究光照对姜黄素诱导人乳腺癌MCF-7细胞凋亡的影响,探讨其作为肿瘤光动力学治疗新型光敏剂的可能性。方法体外培养的MCF-7细胞加姜黄素后进行光照及避光处理24h,MTT法测细胞生长抑制率,Giemsa、Hoechst33258染色观察形态变化,流式细胞术检测凋亡率和细胞周期分布。结果单纯光照对MCF-7细胞无生长抑制及诱导凋亡作用,光照及避光条件下姜黄素对MCF-7细胞生长抑制的IC50分别为2.68、13.44μmol/L;细胞在光照条件下经5μmol/L姜黄素处理24h即呈现典型的凋亡形态;光照条件下1、2.5、3、5、10μmol/L姜黄素及避光条件下5、10、15、20、30μmol/L姜黄素处理细胞24h,凋亡率分别为4.06%、16.79%、19.46%、35.97%、59.27及5.71%、8.88%、12.51%、38.04%、26.60%;姜黄素在光照及避光条件下均可引起细胞周期分布发生改变。结论光照对姜黄素抑制MCF-7细胞生长及诱导凋亡具有增敏作用。

【Abstract】 Objective To investigate the effect of illumination on apoptosis of human breast cancer(MCF-7) cells evoked by curcumine.Methods MCF-7 cells were incubated with or without illumination for 24 hours after addition of curcumine. Cell growth,morphology and apoptosis were determined by MTT,Giemsa and Hoechst33258 staining,and flow cytometry,respectively.Results There was no effect of single illumination on the growth and apoptosis of MCF-7 cells.The IC50 values of curcumine on MCF-7 cells were 2.68 and 13.44 μmol/L with or without illumination,respectively.The typical apoptosis was observed after 24 h exposure to curcumine(5 μmol/L) and illumination.Apoptosis rates of MCF-7 cells treated by curcumine at doses of 1,2.5,3,5 and 10 μmol/L and illumination were 4.06%,16.79%,19.46%,35.97%,59.27%,but those treated only by curcumine at doses of 5,10,15,20 and 30 μmol/L were 5.71%,8.88%,12.51%,38.04% and 26.60%, respectively.Pretreatment with curcumine induced the change of cell cycles regardless of illumination.Conclusion Illumination can reinforce the inhibition and apoptosis of MCF-7 cells caused by curcumine.
