壳核型n-HA/ZrO2支架材料对L929细胞增殖活性的影响
The Effect of Cytoactivity of nano-HA Coating ZrO2 Scaffold on L929 Cells
【摘要】 目的研究核壳型纳米羟基磷灰石包覆二氧化锆(壳核型n-HA/ZrO2)复合生物陶瓷支架材料对L929细胞增殖活性的影响及细胞毒性反应。方法将L929细胞接种于96孔板,随机分为3组进行培养,实验组用壳核型n-HA/ZrO2支架材料浸提液处理,阳性对照组用苯酚溶液,阴性对照组用聚乙烯浸提液,于24、48、72h后观察细胞生长形态,通过MTT比色法检测各组细胞的相对增殖率(relative growth rate,RGR),按GB/T16175-1996标准评价材料的细胞毒性。结果实验组L929细胞24、48、72h后的RGR随时间延长而升高,分别为95.86%、98.29%和102.73%,与阴性对照组L929细胞在增殖活性方面的差异无统计学意义,两组受试材料细胞毒性评级为0~I级。结论核壳型n-HA/ZrO2支架材料不影响L929细胞的增殖活性,无细胞毒性。
【Abstract】 Objective To evaluate the effect of cytoactivity of nano-HA coating ZrO2 scaffold in vitro with MTT assay. Methods According to the standard of GB/T16175-1996,the effects on proliferation of L929 were observed in vitro cultured with the leach liquor of scaffold. The relative growth rate(RGR) was calculated and the cyctotoxicity was graded by a generally accepted standard. Results The RGR of L929 in scaffold group increased gradually from 95.86% in the first day,98.29% in the second day,and 102.73% in the third day,there is significant difference on cell cytoactivity compared with positive control group at the same observed period (P<0.001); and no significant difference compared with negative control group (P>0.05). The grade of cytotoxicity of the scaffold is from 0-I. Conclusion Nano-HA coating ZrO2 scaffold has no evidence of cytotoxicity.
