虎头兰组培快繁技术体系研究
Rapid propagation techniques of Cymbidium hookerianum Rchb
【摘要】 通过对虎头兰无胚乳种子无菌萌发过程中原球茎诱导、继代增殖、试管苗生根等不同发育时期的培养基配方筛选研究,初步建立了一套组培快繁技术体系,结果表明:虎头兰无胚乳种子在温度(25±2)℃、光照强度1 500~2 000 lx、光照时间12 h/d的条件下,较适宜的原球茎诱导培养基为MS+6-BA1.0 mg/L+NAA0.5 mg/L,适宜的增殖培养基为MS+6-BA2.0 mg/L+NAA0.5 mg/L,适宜的生根培养基为1/2MS+NAA1.0 mg/L。
【Abstract】 Based on the reseach of different mediums on fresh bulb inducement,mutiplication,and tube sprout process of no endosperm seeds sterile germination,we establish a rapid propagation system of Cymbidium hookerianum Rchb.The final results shows,the best culture medium of orchid fresh archibulb is MS+6-BA1.0 mg/L+ NAA0.5 mg/L while temprature(25±2)℃ and light 1 500~2 000 lx,12 h/d in no endosperm seeds of Cymbidium hookerianum Rchb,the best medium of mutiplication is MS+ 6-BA2.0 mg/L +NAA0.5 mg/L,and the best medium of rhizogenesis is 1/2MS+NAA1.0 mg/L.
