大豆MIKC型MADS-box基因家族分析

【摘要】 根据拟南芥的MIKC型MADS-box蛋白序列构建HMM(hidden markov model)模型,在大豆基因组中确定了57个MIKC型MADS-box同源基因。分子进化树分析表明,57个大豆MIKC型MADS-box基因分为14个亚类,而MIKC*型基因又分为两个进化支。控制花形态建成的ABCDE同源异型基因(如AP1,AG,AP3和PI等)和重要的控制开花时间的基因(如SVP和SOC1)在大豆基因组中表现出多拷贝的特征。MEME和SMART分析表明,大豆和拟南芥MIKC型MADS-box基因具有保守的MADS和K-box基序,还有一些亚类具有特异的基序。MIKC型MADS-box基因结构较为保守,多数基因具有7~8个外显子。本研究得到的MIKC型MADS-box基因为下一步研究大豆花形态建成和开花时间调控的分子机制提供了重要参考依据。

【Abstract】 MIKC-type MADS-box genes encode a type of transcription factor that play crucial roles in plant development and signal transduction. In the present study, HMM (hidden markov model) profile was employed to identify the MIKC-type MADS-box genes in soybean genome and up to 57 MIKC-type genes were retrieved. Phylogenetic analysis indicated that 57 soybean genes were clustered into 14 subfamilies with their (Arabidopsis thaliana) counterparts and MIKC*-type genes were further clustered into 2 sub-groups. Some floral homeotic gene involved in the ABCDE model for floral development, such as AP1 (apetalal), AG (agamous) and PI (pistilata), and flowering time control genes, such as SVP (short vegetative phase) and SOC1 (suppressor of overexpression of CO 1) were presented as multiple copies. The conserved motifs of MIKC-type MADS-box genes in soybean were identified by MEME and annotated by SMART search program. MADS motif and K-box motif all present in soybean and Arabidopsis MIKC-type MADS-box proteins, and the other sub-groups have their own specific motifs. The exon-intron structure of MIKC-type MADS-box genes is conserved between soybean and Arabidopsis, and most of the genes possess seven or eight exons. The MIKC-type MADS-box genes identified in this study are the good candidate genes for further functional analysis of floral morphogenesis and flowering time control in soybean.
