水稻长链脂肪醇氧化酶基因OsFAO结构分析及其进化研究

【摘要】 我们通过多年来的研究已证实在高等植物中也存在编码类似于工业酵母中长链脂肪醇氧化酶(FAO)的基因。本文对单子叶植物水稻FAO基因的结构、起源和进化进行了分析。利用拟南芥中FAO蛋白的氨基酸序列对水稻基因组进行tblastn及blastp搜索,结果显示在水稻基因组中存在4个FAO基因,1个位于2号染色体,命名为OsFAO1;另外3个在10号染色体上,分别命名为OsFAO2、OsFAO3和OsFAO4。其中位于2号染色体上的OsFAO1基因由3个外显子和2个内含子组成;10号染色体上的3个FAO基因中,OsFAO3和OsFAO4为2个外显子和1个内含子组成,OsFAO2是由4个外显子和3个内含子组成。序列预测结果显示,OsFAO1编码跨膜蛋白的可能性更大。我们进一步对已知序列的各类原核(874种)和各类真核(包括20种真菌)的基因组进行了分析,发现FAO基因主要存在于真菌和陆生植物基因组中,而不存在于原核和藻类的基因组中。对陆生植物基因组中的FAO基因的分析结果表明水稻中OsFAO1基因序列相对原始。

【Abstract】 In previous study, we have identified that in higher plants there exist ω-long chain fatty alcohol oxidase (FAO), which function is similar to those found in industrial yeasts. In this study, we have conducted detail anal- ysis on the gene structure, origin and evolution of the genes encoding FAO in rice (Oryza sativa). The rice genome was searched against the amino acid sequences of 4 FAOs in Arabidopsis by tblastn and blastp. We show that there are four FAO genes in rice genome. One is located at the chromosome 2 and it is designated as OsFAO1. The other three which are named OsFAO2, OsFAO3 and OsFAO4 respectively are all at the chromosome 10. The OsFAO1 gene which is located at the chromosome 2 is composed of 3 exons and 2 introns. Two of the three FAO genes located at the chromosome 10 which are named OsFAO3 and OsFAO4 are composed of 2 exons and 1 introns, the OsFAO2 is composed of 4 exons and 3 introns. Sequence analysis indicate that the OsFAO1 encodes a transmembrane protein. To investigate the origin of the FAO gene, we searched for the FAO sequences in 874 genomes among various prokaryote and eukaryote (including 20 fungi) which have known chromosome sequence. The results show clearly that FAO genes mainly exist in fungal and land plant genomes, but not in prokaryotic and algal genomes. Analysis of four FAO genes in rice indicate that the FAO gene at chromosome 2 is more primitive than the others.
