自噬在大鼠坐骨神经横切后施万细胞去分化中的作用

【摘要】 目的探讨坐骨神经横切后施万细胞去分化的机制。方法建立大鼠坐骨神经损伤模型,实验组隔天腹腔注射自噬抑制剂Ly294002,对照组注射Ly294002的溶剂,于横切后4、7、10 d取坐骨神经近侧端,用免疫组化方法检测施万细胞特异蛋白S100的表达,在电子显微镜下观察施万细胞的形态学变化。结果横切后4 d实验组S100的表达与对照组相比较有显著性差异;S100在幼稚的施万细胞表达很少,随着施万细胞的进一步成熟表达逐渐增多;电子显微镜下观察到施万细胞内有显著的自噬现象。结论自噬在施万细胞清除髓鞘碎片、受损的细胞器和胞质,从而使细胞在重返幼稚状态(去分化)中起重要作用。

【Abstract】 Objective To explore the dedifferentiation mechanism of schwann cells(Scs) after transection of sciatic nerve.Methods Rat models of sciatic nerve injury were established by transecting the left side of rat sciatic nerve.The experiment group were injected abdominally with LY294002(inhibitor of autophagy) every other day,and the control group with its solvent.The expression of S-100 was detected with immunohistochemical method and the morphological changes of Scs were observed by eletron microscope at 4th,7th and 10th day after transection.Results The expression of S-100 in experiment group increased significantly compared with that in control group at 4th day after transection.The expression of S-100 was little in immature Scs,and gradually increased as the cells became more mature.The process of autophagy was observed significantly in Scs.Conclusion Autophagy may play important roles in the dedifferentiation of Scs.
