白藜芦醇抑制人晶状体上皮细胞增殖的信号转导机制
Signal Transduction Mechanism of Resveratrol in Inhibiting Proliferation of Human Lens Epithelial Cell Induced by Recombinant Human Basic Fibroblast Growth Factor
【摘要】 目的探讨白藜芦醇(Res)对重组人碱性成纤维细胞生长因子(rhbFGF)诱导的人晶状体上皮细胞(HLEC)增殖的抑制作用,并从信号转导方面研究其抑制HLEC增殖的作用机制,为寻求防治后发性白内障的有效药物提供实验依据。方法釆用rhbFGF诱导体外培养的HLEC增殖,流式细胞术(FCM)检测一定浓度的Res对增殖状态的HLEC增殖细胞核抗原(PCNA)蛋白表达的抑制作用;FCM检测不同的细胞内信号转导阻断剂(钙调素抑制剂W7、蛋白激酶C抑制剂H7)对其抑制作用的影响。结果Res对HLEC内PCNA蛋白表达有明显的下调作用(P<0.01);W7、H7均明显上调了HLEC内PCNA蛋白表达(P<0.01)。结论Res有抑制HLEC增殖的作用,其机制可能是通过CaM、PKC等来介导的。
【Abstract】 Objective To investigate the proliferative inhibition of human lens epithelial cell(HLEC) induced by recombinant human basic fibroblast growth factor(rhbFGF) by resveratrol(Res) and further study the mechanism of signal transduction in order to provide scientific basis for pursuing safe and effective natural drugs to prevent and cure after cataract.Methods The proliferation of HLEC cultured in vitro were induced by rhbFGF,the inhibition of a certain concentration of Res on the expression of proliferat-ing cell nuclear antigen(PCNA) in proliferative HLEC were detected by flow cytometry(FCM),and the effects of different intracellular signal transduction inhibitor(ie.calmodulin inhibitor W7,protein kinase C inhibitor H7) on its inhibition were detected by FCM.Results Res could down-regulate the expression of PCNA in HLEC significantly(P<0.01).W7 and H7 could up-regulate the expression of PCNA in HLEC significantly(P<0.01).Conclusion Res can inhibit the proliferation of HLEC induced by rhbFGF,and the mechanism may be related to CaM and PKC signal transduction pathway.
