川芎嗪对糖基化终末产物诱导肾小管上皮细胞外基质表达的影响
Effects of Ligustrazine on Expression of Extracellular Matrix in Human Proximal Renal Tubular Epithelial Cells Induced by Advanced Glycation End Products
【摘要】 目的观察川芎嗪对AGE-BSA诱导人近端肾小管上皮细胞(HK-2)细胞外基质(ECM)成分表达的影响,探讨其干预糖尿病肾间质纤维化的作用机制。方法将HK-2细胞在体外与BSA、AGE-BSA、AGE-BSA+川芎嗪(不同浓度)共培养72h后,用RT-PCR和Western Blot分别检测FN、ColⅠα1mRNA及蛋白的表达。结果RT-PCR和Western Blot结果显示,100μg/ml BSA刺激HK-2细胞后,FN、ColⅠα1mRNA及蛋白表达无明显增加;100μg/ml AGE-BSA刺激后,FN、ColⅠα1mRNA及蛋白表达明显增加,与空白对照组及相同浓度BSA组相比,P<0.01;加入(40~120)μg/ml川芎嗪共同干预后,川芎嗪可在mRNA及蛋白水平呈剂量依赖性下调AGE-BSA诱导的FN、ColⅠα1的表达。结论川芎嗪能抑制AGE-BSA诱导的HK-2细胞FN、ColⅠα1的合成。表明川芎嗪能通过抑制肾小管上皮细胞分泌ECM进而延缓糖尿病肾间质纤维化的进展。
【Abstract】 Objective To investigate the effect of ligustrazine on the expression of extracellular matrix(ECM) components in human renal proximal tubular cells(HK-2) induced by advanced glycation end products(AGEs) ,and explore it’s interventional mechanism on tubulointerstitial fibrosis in diabetic nephropathy. Methods HK-2 cells were cultured in vitro with bovine serum albumin(BSA) ,AGE-BSA and recombinant AGE-BSA and ligustrazine respectively for 72 h. The expressions of FN and ColⅠα1 mRNA and protein were detected by RT-PCR and western blot respectively. Results The expression levels of FN and Col Ⅰα1 mRNA and protein were up-regulated induced by 100 μg/ml AGE-BSA compared with the blank control group and the same concentration group of BSA(P<0.01) . 40~120 μg/ml ligustrazine was added to the culture solution,the expression levels induced by AGE-BSA were dramatically down-regulated in a dose-dependent manner. Conclusion Ligustrazine negatively regulates the synthesis of FN and ColⅠα1 induced by AGE-BSA. This shows that ligustrazine can delay the development of tubulointerstitial fibrosis in diabetic nephropathy by inhibiting the accumulation of ECM in HK-2.
