【摘要】 目的本研究分析Stat3及其靶基因产物cyclin D1与Bcl-xl在喉癌组织与细胞中的表达情况,探讨Stat3及其靶基因产物在喉癌发病机制中的作用。方法用Westernblot检测50例喉癌组织、10例喉癌旁黏膜以及喉癌细胞系Hep-2中Stat3、p-Stat3及其靶基因产物Cyclin D1和Bcl-xl蛋白表达,用免疫组织化学染色进行细胞内定位,同时对喉癌组织中p-Stat3、Cyclin D1及Bcl-xl表达情况与不同临床病理特征进行统计学分析。结果喉癌组织中Stat3、p-Stat3、Cyclin D1及Bcl-xl蛋白表达水平(A值)分别为86.97±1.58、56.97±3.93、24.43±5.62、33.78±3.56;癌旁黏膜分别为36.10±0.526、16.52±0.529、5.94±1.75、14.68±2.14)P<0.05)。p-Stat3表达水平与喉癌临床分期、病理分级及淋巴结转移有关(P=0.026、0.019、0.034);Cyclin D1表达水平与临床分期、病理分级、淋巴结转移(P=0.015、0.027、0.034)有关;Bcl-xl表达水平与临床分期、淋巴结转移(P=0.023、0.033)有关。p-Stat3与Cyclin D1表达呈线性相关(r=0.451,P<0.05)。结论Stat3信号转导通路可能在喉癌的发生发展过程中起重要作用,检测喉癌中Stat3及其靶基因产物的表达可以反映肿瘤的恶性程度和病情进展。更多还原

【Abstract】 OBJECTIVE This study was conducted to investigate the expression of Stat3 and its target gene products including Cyclin D1 and Bcl-xl in human laryngeal cancerous tissue, and to explore the mechanism in tumorigenesis of larymgeal carcinoma, and to explore the correlation between the expression,clinical parameters as well as the pathological parameters of laryngeal carcinoma. METHODS The expression of Stat3, p-Stat3, Cyclin D1 and Bcl- xl in 50 cases of cancerous tissues, adjacent normal tissues was measured by Western blot analysis. The expression pattern of Stat3 and its activated form p-Stat3 was determined by immunohisto- chemical staining. The correlation of the expression of Stat3, p-Stat3, Cyclin D1 and Bcl-xl in laryngeal carcinoma with various clinicopathological characteristics was analyzed statistically. RESULTS The protein expression level of stat3, p-Stat3, Cyclin D1, Bcl-xl (A value)were 86.97±1.58, 56.97±3.93, 24.43±5.62, 33.78± 3.56 in laryngeal carcinoma and 36.10±0.52, 16.52±0.52, 5.94±1.75, 14.68±2.14 in adjacent normal respectively(P<0.05). Over- expression of p-Stat3, Cyclin D1 was correlated with clinical stage(P=0.026, 0.015), histological grade(P=0.019,0.027)and lymph node metastases(P=0.034, 0.034). Elevated levels of Bcl-xl were associated with clinical stage (P=0.023)and lymph node metastases(P=0.033). It was found that Cyclin D1 was in a positive linear correlation fashion with p-Stat3 in tumor(r=0.451,P<0.05). CONCLUSION Stat3 signaling pathway may play an important role in the tumorigenesis of laryngeal carcinoma and disease progression. Determination of Stat3 and its target gene products can be used to indicate the malignancy degree of laryngeal carcinoma.更多还原
