电镜重构病毒的三维显示与三维任意区域分割
Three-dimensional representation and arbitrary region segmentation in 3D reconstruction of virus particles by electron cryo-microscopy
【摘要】 利用冷冻电镜单颗粒三维重建方法获得分辨率1.5nm家蚕质多角体(BmCPV);2.5nm,中蜂囊状幼虫病病毒(CSBV);1.4nm,伊蚊C6/36细胞浓核病毒(C6/36DNV)的三维结构体数据,对三维结构数据进行了数据归并,并找到对显示细节较为敏感的向量维度,用表面显示方法对病毒体数据进行三维显示,用基于空间的方法完成了任意区域的交互分割。显示时间和信噪比与原有显示方法有了提高。病毒表面细节和轴上突起清晰可见。所分割的部分结构准确完整,并可以对所分割的部分进行量化分析。为冷冻电镜单颗粒重构提供了新的三维可视与分析方法。
【Abstract】 Recent advances in electron cryomicroscopic instrumentation and single particle reconstruction method have created opportunities for high-throughput and high-resolution three-dimensional(3D)structure determination of virus.In order to visual and understand effectively these 3D structure,here,we present a display method based on surface rendering with the function of 3D arbitrary region interactive segmentation and quantitative analysis,and integration them into software package CEM-3DDS(electron cryo-microscopy three-dimensional virus display and arbitrary region segmentation system).CEM-3DVDDSS consists of a complete set of modular programs for icosahedral virus 3D representation and segmentation organized under a graphical user interface and provides options for user-friendly.Firstly,we translate volume data form format MRC obtaining from electron cryo-microscopy single particle reconstruction to format fitting our software.In the processing,the original volume data is merged for compression data and a better vector dimension is found for controlling detail resolution in the data translation.Then,the new volume data can be displayed and segmented using CEM-3DVDDSS.We demonstrate the applicability of CEM-3DVDDSS by using it to display 1.5nm(resolution)BmCPV(Bombyx mori Cytoplasmic Polyhedrosis Virus),2.5 nmCSBV(Chinese Sacbrood Bee Virus),and 1.4 nm C6/36DNV(Bensonucleosis Virus)3D structure.As a results,both 3D display speed and signal to noise ratio are improved comparing to original method,and segmentation results are exact and whole with additive function of quantitative analysis to 3D structure.
