黄芪注射液对TNF-α诱导人脐静脉内皮细胞炎症因子的影响及其分子机制
Effects of Astragalus Membranaceus Injection on TNF-α-induced release of inflammatory factors from HUVECs and the molecular mechanisms
【摘要】 目的观察黄芪注射液对肿瘤坏死因子-α(TNF-α)诱导的体外培养人脐静脉血管内皮细胞(HUVECs)核因子-κβ(NF-κβ)激活与白介素-6(IL-6)、可溶性细胞间黏附分子-1(sICAM-1)分泌的影响,探讨其分子机制。方法体外培养的第2代HUVECs用于实验,免疫组织化学分析检测细胞NF-κβ亚单位p65的表达程度,Western blot检测核内NF-κβ含量变化,ELISA法检测培养液中IL-6、ICAM-1浓度。结果TNF-α有效激活NF-κβ并诱导HUVECs分泌IL-6、sICAM-1增加;黄芪注射液预先孵育2h能抑制NF-κβ表达并减轻TNF-α诱导的上述反应。结论TNF-α可能通过激活NF-κβ使IL-6、sICAM-1增加,从而损伤血管内皮功能;黄芪注射液对上述反应有拮抗作用,推测是通过抑制NF-κβ途径实现血管内皮保护功能。
【Abstract】 Objective To investigate the protective effect of Astragalus Membranaceus Injection on human umbilical vein endothelial cells (HUVECs) against tumor necrosis factor α (TNF-α). Methods Cultured passage 2 HUVECs were stimulated with TNF-α with or without a 2-h Astragalus Membranaceus Injection treatment. The expression of nuclear factor-κB (NF-κB) subunit p65 were evaluated by immuncytochemistical method, and the levels of p65 in the nuclei and the protein Iκβα in the cytoplasm were evaluated by Western blotting. The levels of interleukin-6 (IL-6) and soluble intracellular adhesion molecule-1 (sICAM-1) in the cell culture were determined with ELISA. Results TNF-α induced the activation of NF-κB and increased the expressions of IL-6 and sICAM-1 in HUVECs. The activation of NF-κβ by TNF-α was suppressed by Astragalus Membranaceus Injection in a dose-dependent manner. Conclusion Astragalus Membranaceus Injection can inhibit the TNF-α-induced expression of IL-6 and sICAM-1 by suppressing NF-κβ activation, suggesting its protective effect on the endothelial function.
