局部制动对大鼠胰岛素敏感性影响的研究

【摘要】 目的:了解肢体制动对大鼠胰岛素敏感性的影响.方法:将SD大鼠随机分为对照组、制动组,每组20只.制动组大鼠右后肢石膏固定,对照组不做特殊处理.3wk后将两组大鼠行高胰岛素—正常葡萄糖钳夹技术,术后分离大鼠右比目鱼肌测量质量、肌细胞直径及肌纤维截面积.结果:制动组稳态葡萄糖输入速率[(35±5)mg(/kg·h)vs(13±4)mg/(kg.h)]、右侧比目鱼肌湿质量[(169.2±4.1)gvs(67.1±6.3)g]、肌细胞直径[(49.8±2.7)μmvs(20.6±3.2)μm]、肌纤维截面积[(998±21)μm2vs(579±14)μm2]均明显低于对照组.结论:局部制动可导致大鼠骨骼肌萎缩,并使机体对胰岛素的敏感性降低.

【Abstract】 AIM:To observe the effects of limb immobilization on the insulin sensitivity.METHODS:Sprague-Dawley rats were randomly assigned to 2 groups:control(CON)and three-week immobilization(IB).The right legs of the rats in IB group were immobile for three weeks.After the 3 weeks,hyperinsulinem ic-euglycemic clamp was used to evaluate the insulin sensitivity and soleus muscles were dissected and weighted.The diameter and cross section area of muscle cell were measured.RESULTS:In IB group,the wet weights of the soleus [(169.2±4.1)g vs(67.1±6.3)g],the diameter of the muscle cell [(49.8±2.7)μm vs(20.6±3.2)μm)] and the area of its cross section decreased [(998±21)μm2 vs(579±14)μm2)] obviously as compared with those in CON group.The glucose infusion rate was significant lower than that in CON group [(35±5)mg/(kg·h)vs(13±4)mg/(kg·h)].CONCLUSION:Limb immobilization induces soleus atrophy and decreases the insulin sensitivity.
