用热休克法诱导虹鳟雌核发育二倍体的研究
Diploid gynogenesis induced by heat shock in rainbow trout
【摘要】 采用经紫外线照射后遗传上失活的精子激活卵子,用热休克法诱导虹鳟Oncorhynchus mykiss减数分裂雌核发育二倍体。对影响诱导效果的紫外线照射精子的剂量及28℃热休克处理的持续时间和处理起始时刻进行了研究。结果表明:紫外线照射精子的最佳条件为照射距离8 cm、照度6 670μW/cm2、照射时间15 min;获得虹鳟减数分裂雌核发育二倍体的最佳参数为受精后20 min,28℃热休克处理10、15 min,雌核发育率为14.28%~37.75%。
【Abstract】 Meiotic diploid gynogenetic fry of rainbow trout,Oncorhynchus mykiss,were produced from the embryos activated by UV-irradiated sperm with heat shock.The UV radiation levels,and duration and application time of 28 ℃ heat shock were investigated.It was found that the best results were observed at UV radiation distance of 8cm,and the corresponding level of 6 670 μW/cm2 for 15 min.The 10 min and 15 min treatments 20 min after activation with 28 ℃ heat shock led to the highest yields of diploid gynogens.The rate of gynogenetic induction ranged from 14.28 to 37.75%.
