高效液相色谱法测定人脑脊液中左氧氟沙星的方法学研究

【摘要】 目的:建立脑脊液中左氧氟沙星含量的RP-HPLC测定方法。方法:采用Shim-pack VP-ODS C18色谱柱(150mm×4.5mm,5μm);乙腈-0.1%甲酸(三乙胺调PH2.52)=16:84为流动相,用前经0.45μm的滤膜过滤,超声脱气;流速为1.0ml/min;紫外检测波长为297nm;柱温为30℃,进样量20μl。结果:此方法最低检测浓度为24.19ng/ml,线性范围为24.19~6200ng/ml,方法平均回收率在96.03%~102.08%,日内及日间精密度的相对标准差分别为2.64%~9.12%和2.59%~8.38%。结论:该方法简单、快速、灵敏度高,重复性好,可用于左氧氟沙星的脑脊液浓度检测测。

【Abstract】 Objective: To determine levofloxacin in the human cerebrospinal fluid by revers-ed phase high performance liquid chromatography. Methods: Levofloxacin was successfully sep-arated using a Shim-pack VP-ODS C18 column(150mm×4.6mm, 5μm). The mobile phase consist-ed of acetonitrile-0.1% formic acid solution (16:84) (adjusted the PH to 2.52 with triethy-lamine) and was degassod ultrasonically before use. Each component of the mobile phase was filtered through a 0.45μm membrane. The column temperature was 30℃ and the flow rate was 1.0ml/min. Injection volume was 20μl. The detection wavelength was 297nm. Results: The de-tection limit was 24.19ng/ml. The linear range was 24.19~6200ng/ml. The average recovery was 96.03%~102.08%. The RSD for intra-day and inter-day was 2.64%~9.12% and 2.59%~8.38%, re-spectively. Conclusions: The method is simple, rapid, sensitive, producible and can be used to monitor the concentration of levofloxacin in the patients cerebrospinal fluid.
