栀子组培苗的瓶外生根

【摘要】 为了降低栀子组培苗生产成本,利用无土栽培技术进行了组培苗的瓶外生根培养。结果表明:以薄泡沫板做为支持物,营养液pH值为5.0、电导率为1.5×10-3S.cm-1时有利于幼苗生根及生长。对瓶外生根苗与组培苗和温室植株叶片的气孔数及气孔形态进行观察比较的结果表明,瓶外生根培养7 d苗的叶片气孔与组培苗相似,数量多且较大;而瓶外生根培养14 d苗的叶片气孔与温室1年生苗相似,几乎所有气孔都恢复开闭功能。瓶外生根苗移植于河沙和有机肥混合基质时,成活率达到100%,且长势强,新叶生长情况良好。

【Abstract】 An experiment was conducted to study the ex vitro rooting of micropropagated shoots in Gardenia jasminoides in order to decrease production cost of plantlet in tissue culture by using the method of soilless culture.The effects of culture supporter,pH and electrical conductivity(EC) of nutrient solution on rooting and growth of G.jasminoides were investigated.The optimal rooting and growth conditions for G.jasminoides were obtained as pH 5 and EC 1.5×10-3S·cm-1 using foam as supporter.The stomatal number per leaf and stomatal morphology for ex vitro culture were compared with those for in vitro culture and greenhouse culture.Stomatal number and stomatal size of ex vitro plantlets were similar to those of in vitro ones after cultivation for 7 days,and a number of large stomata were observed.However,the stomata of ex vitro plantlets were correspondent with those of one-year-old plantlet in greenhouse after cultivation for 14 days,and almost all the stomata were found to recover the open/close function.Moreover,the survival rate of ex vitro plantlets transplanted to the mixture substrate of sand and organic fertilizer could reach 100%,and the plantlets with well-developed new leaves grew very well.
