黄芪多糖对仿刺参体腔细胞中溶菌酶基因表达量的影响

【摘要】 为研究黄芪多糖诱导仿刺参体腔细胞中溶菌酶基因表达的影响,从体质量(10±1.22)g的仿刺参体腔细胞中克隆出长度为395 bp的-βactin基因部分片段作为内参,利用半定量PCR法考察体内注射不同剂量黄芪多糖(APS)后仿刺参体腔细胞中i型溶菌酶mRNA表达量变化。研究发现,5、10mg/kg剂量的APS可提高体细胞中溶菌酶mRNA表达量,其中5 mg/kg剂量组达显著水平,剂量为20 mg/kg时基本无效。

【Abstract】 Effects of astragalus polysaccharides(APS) on gene expression of lysozyme in coelmocytes of sea cucumber Apostichopus japonicus [mean body weight:(10±1.22) g] were studied.First,a 395 bp β-actin gene fragment was amplified from coelmocytes for reference.The expression of lysozyme mRNA was determined using semiquantitative reverse transcription-polymerase chain reaction(RT-PCR) analysis in vitro.Results showed that lysozyme mRNA level increased at a dose of 5 mg/kg and 10 mg/kg.A significant increase was observed at 5 mg/kg,and there was no significant effect of high dose of APS(20 mg/kg) on lysozyme mRNA expression.
