人趋化素样因子1对人动脉血管平滑肌细胞的趋化作用

【摘要】 目的:观察人趋化素样因子1(chemokine-like factor1,CKLF1)对人外周动脉血管平滑肌细胞(arterial smooth muscle cells,ASMCs)的趋化作用。方法:收集用CKLF1真核细胞表达载体(pEGFP-N1-CKLF1)瞬时转染293T细胞72h后的上清培养液,采用细胞迁移实验检测CKLF1对ASMCs的趋化作用。结果:未稀释和10倍稀释的上清培养液孵育后的实验组迁移细胞数与对照组(pEGFP-N1)迁移细胞数相比,差异有统计学意义(114±4vs41±4,P<0.05;74±4vs34±3,P<0.01)。而100倍和1000倍稀释的上清培养液孵育后实验组迁移细胞数和对照组的迁移细胞数相比,差异无统计学意义(28±4vs25±5,P>0.05;26±5vs23±5,P>0.05)。ASMCs分别被不同浓度百日咳毒素(pertussis toxin,PTX)处理后,PTX浓度为0和2μg/L时,10倍稀释的上清培养液实验组迁移细胞数与对照组迁移细胞数相比,差异有统计学意义(74±4vs34±3,P<0.01;45±3vs34±3,P<0.01)。PTX浓度为10μg/L,10倍稀释的上清培养液实验组迁移细胞数与对照组迁移细胞数相比,差异无统计学意义(37±4vs34±3,P>0.05)。结论:CKLF1对人动脉平滑肌细胞具有明显趋化作用,且该趋化作用可被PTX所阻断。

【Abstract】 Objective:To investigate the chemotactic effect of chemokine-like factor 1(CKLF1)on human arterial smooth muscle cells(ASMCs).Methods:The recombinant eukaryotic expression vectors pEGFP-N1-CKLF1(test group) and pEGFP-N1(control group)were transiently transfected into 293T cells.The supernatants were harvested 72 h after transfection for bioactivity study.The chemotactic effect of CKLF1 on ASMCs were assayed by cellular chemotactic experiments.Results:ASMCs number migrated from the test and control groups diluted by none and 10-fold supernatants had statistical significance(114±4 vs 41±4,P<0.05;74±4 vs 34±3,P<0.01),but have no statistical significance(28±4 vs 25±5,P>0.05;26±5 vs 23±5,P>0.05) between the two groups diluted by 100-fold and 1 000-fold supernatants.When ASMCs were treated at different concentrations of 0 and 2 μg/L of pertussis toxin(PTX),the cell number migrated from the test and control groups diluted by 10-fold supernatants,they had statistical significance(74±4 vs 34±3,P<0.01;45±3 vs 34±3,P<0.01).And when ASMCs were treated at 10 μg/L of PTX,the cell number migrated from both groups diluted by 10-fold supernatants,they had no statistical significance(37±4 vs 34±3,P>0.05).Conclusion:CKLF1 has significant chemotactic effects on ASMCs and such a CKLF1-induced chemotaxis could be inhibiteded by PTX at concentration of 10 μg/L.
