过逸气虚证候动物模型的建立方法

【摘要】 目的:建立过逸气虚证候大鼠模型;方法:60只雄性Wistar大鼠随机分为空白对照组、气虚模型组、人参组、薤白组,每组15只。空白对照组大鼠置于普通代谢笼中(单笼),饲以普通饲料;气虚模型组大鼠置于经改造过的代谢笼中(单笼),笼内空间以其保持基本安静且能转身移动为标准,饲以高营养饲料;人参组与薤白组大鼠于实验开始分别给予人参(1.2g生药.kg-1.d-1)与薤白(1.2g生药.kg-1.d-1)CMC-Na混悬溶液灌胃,1次/d,其他处理同气虚模型组。共计10周。实验结束后对大鼠生物学表征进行观察,并采用半定量评分表评分;测定大鼠力竭游泳时间。结果:与空白对照组比较,气虚模型组大鼠生物学表征明显改变,评分显著升高(P<0.01),力竭游泳时间显著缩短(P<0.01);与气虚模型组比较,人参组大鼠生物学表征明显改善,评分减少(P<0.05),力竭游泳时间显著延长(P<0.01);薤白组上述指标无显著性差异。结论:采用高营养饮食复合限制活动的方法可成功建立气虚证候动物模型。

【Abstract】 Objective: To establish rat model of qi deficiency syndrome caused by excessive idleness. Methods: 60 male Wistar rats were divided into 4 groups randomly: blank control group, qi deficiency group,Ginseng group and Bulbus allii macrostemi group. The rats of blank control group were put into single caves and fed with common feedstuff; the rats of qi deficiency group were put into the remoulded single caves, the standard is that the rats can keep basic quiet and can turn round in the space, the rats were fed with hyperalimentation feedstuff; the rats of Ginseng group and Bulbus allii macrostemi group were given ginseng(1.2g crude drug /kg/d) and bulbus allii macrostemi(1.2g crude drug /kg/d) freeze drying powder intragastrically respectively in the beginning of the experiment, one time everyday, other factors are the same as the qi deficiency group. All the rats were fed with 10 weeks continuously. At the end of experiment, the biology superficial syndrome and exhausted swimming time of the rats were observed. Results: Compared with the blank control group, the biology superficial syndrome scores of qi deficiency group increased obviously(P<0.01), exhausted swimming time decreased obviously(P<0.01). Compared with model group, the biology superficial syndrome scores decreased(P<0.05)and exhausted swimming time increased remarkably(P<0.01)in rats of Ginseng group, but above-mentioned items had no changes in Bulbus allii macrostemi group. Conclusion: It can establish animal model of syndrome of qi deficiency caused by excessive idleness successfully by the method of using hyperalimentation feedstuff combined with exercise restraint.
