益气血祛风湿方对阿霉素肾病大鼠肾组织α-SMA、ROCK1表达的影响

【摘要】 目的:探讨益气血祛风湿方治疗肾脏病的有效作用和细胞生物学机制。方法:采用阿霉素肾病大鼠模型,用益气血祛风湿方水煎剂以及黄芪当归和青藤碱等进行干预治疗4周。实验结束时留取肾脏组织,采用免疫组化、Western-blotting技术观察肾组织中α-平滑肌肌动蛋白(α-SMA)的蛋白表达;采用RT-PCR技术测定肾组织中α-SMA、Rho相关卷曲螺旋形成蛋白激酶1(ROCK1)等基因表达水平。结果:模型组大鼠肾组织中α-SMA的蛋白表达较正常大鼠显著增多,α-SMA和ROCK1基因表达也显著增高。各治疗组大鼠肾组织中α-SMA的表达均较模型组显著降低,益气血祛风湿方与青藤碱组显著优于芪归治疗组。各治疗组大鼠肾组织中α-SMA、Rock1基因表达水平较模型组显著降低,治疗组间无显著差异。结论:益气血祛风湿方显著降低肾间质纤维化相关分子的表达,在α-SMA方面作用较黄芪当归显著增强。

【Abstract】 Objective:To observe the effects of YiqixueQufengshiFang and its compositions(Astragali and Angelica,Sinomenine) on renal interstitial fibrosis by examining the expression of α-smooth muscle actin(α-SMA)and Rho associated coiled coil forming protein kinase 1(ROCK1)in kidney tissue of rats with adriamycin nephrosis.Methods:The rat model of adriamycin nephrosis was established by intravenous injection of adriamycin .The rats with nephropathy were treated with YiqixueQufengshiFang,Astragali and Angelica(A&A),or Sinomenine for 4 weeks.The natural rats were regarded as control group.Immunohistochemistry and Western-blotting combined with semi-quantitive method were used to investigate the protein level of α-SMA in renal tissues.The mRNA expression of α-SMA and ROCK1 in renal tissue was measured by RT-PCR.Results :In adriamycin nephrosis rats,the expression level of α-SMA in renal tissue increased significantly.The expression level of α-SMA were alleviated by YiqixueQufengshiFang,A&A,or Sinomenin treatment.YiqixueQufengshiFang and Sinomenin treatment showed a better effect on suppressing the protein expression of α-SMA than A&A.All of treatments significantly suppressed the mRNA expression of α-SMA and ROCK1,but no obvious difference between them.Conclusion:YiqixueQufengshiFang has more efficiency in preventing interstitial fibrosis and suppressing interstitial myofibroblast in renal interstitium than A&A.
