人膜联蛋白的表达及性质初步研究
EXPRESSION AND CHARACTERIZATION OF HUMAN ANNEXIN Ⅴ
【摘要】 基于人膜联蛋白Ⅴ(Annexin Ⅴ)cDNA分子构建了相应的大肠杆菌融合表达体系.通过摸索和优化表达条件,表达并纯化出可溶的GST-重组人Annexin Ⅴ融合蛋白.通过活化部分凝血活酶时间(tAPT)实验测定表明,GST-重组人Annexin Ⅴ融合蛋白能有效延长tAPT,具有较强的抗凝活性,且延长时间与剂量成线性关系.
【Abstract】 Human AnnexinⅤ was expressed as a fusion protein with GST protein in host cell E.coli BL21. Expressed protein was purified to high homogeneity by affinity chromatography. Recombinant GST-Human AnnexinⅤ fusion protein could efficiently prolong activated partial thromboplastin time (APTT) in vitro and suggesting that it has potent anti-coagulant activity.
