氯霉素全抗原的合成与鉴定
Synthesis and Identification of Chloramphenicol Antigen
【摘要】 利用重氮化法合成氯霉素全抗原,将氯霉素的对位苯硝基构造成芳香氨基,并氧化使其与牛血清白蛋白偶联,构成氯霉素全抗原。此外,利用紫外光谱、红外光谱与酶联免疫等方法对合成的全抗原进行鉴定。结果表明,氯霉素全抗原合成成功,并免疫原性良好。其偶联比为1∶0.959,蛋白质浓度达2.799 mg/mL。
【Abstract】 Diazotizationis adopted to synthesize the complete antigen of chloramphenicol(CAP)in this paper.Chloramphenicol was reformed and then was coupled with carrier protein bovine serum albumin(BSA).The completeantigen was identified by ultraviolet spectrum,infrared spectrum and enzyme-linked immunosorbent assay.Results indicated that synthesis of CAP antigen was successful and it had immunogenicity.The coupling ratiowas 1∶0.959 and concentration of protein was 2.799 mg/mL.
