乙型肝炎病毒表面抗原118位氨基酸变异研究

【摘要】 目的研究献血者乙型肝炎病毒表面抗原118位氮基酸变异对ELISA检测的影响。方法PCR扩增献血者表面抗原基因,恢复变异点氨基酸为野生型,构建不同的表达载体,在293T细胞株中表达,检测各自在不同试剂中的反应格局。结果117位点对检测反应格局不产生明显影响,118位点的变异明显影响检测格局,恢复变异点氨基酸明显提高了检出率和灵敏度。结论118位氮基酸变异影响了乙肝表面抗原的筛查,改变了其的免疫反应原性。

【Abstract】 Objective To study the effect of HBsAg polymorphism at aa118 position on the blood HBsAg screening test.Methods HBsAg gene,amplified by PCR,was cloned and sequenced,then mutated amino acids were recovered into the wild type at aa117 or/and aa118 position. By the insertion of HBsAg genes,expression vectors were constructed and transient expressions were committed in 293T cells. Suspensions of cell culture were collected and tested by the panel,which contained 10 kinds of HBsAg commercial testing kits.Result The profile of results tested by Panel was similar between expressing suspension of mutation HBsAg gene and donation serum,and most of kits but two failed. But the profile was significantly different compared with normal quality control serum. When mutation was recovered at 117 position,the profile of results did not change. But mutation was recovered at 118 position,the profile of results changed,and all kits can test this HBsAg effectively.Conclusion Mutation of HBsAg at 118 position,which was changed from T to M,leaded to the failure tests by most of kits.
