低氧训练对大鼠外周血CD34~+、CD34~+/Flk-1~+细胞数量的影响

【摘要】 探讨低氧及运动训练对大鼠外周血CD34+、CD34+/Flk-1+细胞数量的影响。研究方法:健康雄性SD大鼠60只,随机分为6组:对照组、高住组、常练组、低住高练组、高住低练组、高住高练组。采用递增负荷跑台运动训练及低氧作为处理因素,建立大鼠低氧训练模型。采用Ficoll密度梯度离心分离单核细胞,应用流式细胞术检测外周血单个核细胞表型。研究结果:对照组、高住组、低练组、低住高练组、高住低练组、高住高练组大鼠外周血单个核细胞中CD34+细胞百分率分别为0.19%、0.28%、0.75%、2.04%、6.51%、8.39%,高住组、低练组、低住高练组与对照组相比,增加不具显著性,高住低练组、高住高练组与对照组相比显著增加;CD34+/Flk-1+细胞百分率分别为0.03%、0.10%、0.26%、0.65、1.65%、2.42%。高住组、低练组与对照组相比,增加不具显著性,低住高练组、高住低练组、高住高练组与对照组相比显著增加。结论:低住低练可获得较高的CD34+细胞、CD34+/Flk-1+细胞动员率,外周血CD34+细胞数量增加的程度高于CD34+/Flk-1+细胞增加的程度;CD34+/Flk-1+细胞可能作为重要的细胞供体,参加低氧运动心肌组织的血管新生。

【Abstract】 Objective: To study the effect of hypoxia,training and hypoxic training on the numbers of CD34+ cells and CD34+/Flk-1+ cells in rats’ peripheral blood.Materials and Methods: 60 male SD rats are randomly divided into six groups.Progressive treadmill exercise and hypoxia are used to establish hypoxic training animal model.Peripheral blood lymphocytes of rats are separated by Ficoll-Urografin density gradient centrifugation.Flow cytometry is applied to exam phenotype of peripheral blood mononuclear cells.The results show that the percentage of peripheral blood CD34+ cells are 0.19%,0.28%,0.75%,2.04%,6.51%,8.39% in normoxic control group,hypoxic living group,normoxic training group,normoxic living hypoxic training group,hypoxic living normoxic training group,and hypoxic living hypoxic training group respectively.Hypoxic living normoxic training and hypoxic living hypoxic training increase the percentage of peripheral blood CD34+ cells significantly,compared with normoxic control group.The percentage of peripheral blood CD34+/Flk-1+ cells are 0.03%,0.10%,0.26%,0.65%,1.65%,2.42% in normoxic control group,hypoxic living group,normoxic training group,normoxic living hypoxic training,hypoxic living normoxic training,and hypoxic living hypoxic training respectively.Normoxic living hypoxic training,hypoxic living normoxic training,and hypoxic living hypoxic training increase the percentage of peripheral blood CD34+/Flk-1+ cell significantly compared with normoxic control group.Conclusion: Together with hypoxic living and hypoxic training can obtain high CD34+ cell and CD34+/Flk-1+ cell mobilization rates,and CD34+ cell mobilization rate is higher than that of CD34+/Flk-1+ cell.In addition,CD34+/Flk-1+ cell may take part in angiogenesis in cardiac muscle as an cell offer.
