Wnt-1、β-catenin在鼻咽癌组织中的表达及其临床意义
Expression and clinical significance of Wnt-1 and β-catenin in nasopharyngeal carcinoma
【摘要】 背景与目的:Wnt-1和β-catenin分别作为Wnt/β-catenin信号通路中的信号分子及关键分子在多种肿瘤中异常表达,且与预后不良有关。本研究通过检测Wnt-1、β-catenin在鼻咽癌组织中的表达,探讨其与鼻咽癌临床预后的关系。方法:用免疫组化SP法检测111例鼻咽癌组织中Wnt-1、β-catenin蛋白的表达情况,分析其与鼻咽癌无复发生存率、无转移生存率和无进展生存率的关系。结果:111例标本中,β-catenin高表达64例(57.7%),晚期鼻咽癌β-catenin的高表达率高于早期(63.1%vs40.7%,P=0.041);β-catenin高表达组的无复发生存率、无转移生存率、无进展生存率均显著低于低表达组(P<0.05)。多因素Cox回归模型显示β-catenin高表达与肿瘤进展有关。Wnt-1高表达68例(61.3%),高表达组与低表达组的无复发生存率、无转移生存率、无进展生存率相比,两者差异无统计学意义(P>0.05)。结论:部分鼻咽癌中可能存在Wnt/β-catenin通路的异常活化,β-catenin可作为鼻咽癌的一个预后指标。
【Abstract】 Background and Objective: As signaling molecule and key component of Wnt / β-catenin signaling pathway respectively, Wnt-1 and β-catenin are abnormally expressed in several malignancies and correlate with poor prognosis. This study was to investigate the expression and clinical significance of Wnt-1 and β-catenin in nasopharyngeal carcinoma (NPC). Methods: The expression of Wnt-1 and β-catenin in 111 specimens of NPC was detected by SP immunohistochemistry. Their correlations to relapse-free survival (RFS), metastasis-free survival (MFS) and progression-free survival (PFS) were analyzed. Results: The high expression of β-catenin was observed in 64 (57.7%) of the 111 cases. Its high expression rate was significantly higher in advanced NPC than in early stage NPC (63.1% vs. 40.7%, P=0.041). The RFS, MFS and PFS were lower in high β-catenin expression group than in low β-catenin expression group (P<0.05). Cox regression analysis demonstrated that β-catenin was related to poor prognosis of NPC patients. The high expression of Wnt-1 was observed in 68 (61.3%) of the 111 cases, but its expression had no effect on RFS, MFS and PFS (P>0.05). Conclusions: Wnt / β-catenin signaling pathway may be activated abnormally in some NPC patients. β-catenin may be a prognostic factor of NPC.
