重组抗原rMIC3-ELISA检测猪弓形虫抗体的研究
Detection of the antibody against Toxoplasma gondii using the indirect ELISA with the recombinant MIC3
【摘要】 [目的]探讨检测猪弓形虫病的新方法。[方法]应用纯化的重组弓形虫微线体蛋白3(rMIC3)作为包被抗原建立间接ELISA方法,用于猪弓形虫抗体的检测。[结果]该法所用最佳抗原浓度为3.40μg/ml,最适宜的血清稀释倍数为1∶160,与猪瘟病毒等阳性血清不发生交叉反应,与LAT的符合率为92.56%。[结论]该方法快速、特异、重复性好,可用于猪弓形虫病的诊断和流行病学的调查。
【Abstract】 [Objective]The aim of this research was to explore a new method for serodiagnosis of swine toxoplasmosis.[Method]An indirect ELISA with the recombined MIC3 was standardized for detecting the antibody against Toxoplasma gondii.[Result] The optimum working oncentration of the recombinant MIC3 and the serum samples was 3.40 μg /ml and 1∶160 respectively.The cross-reaction between Toxoplasma gondii positive serum and classical swine fever virus positive serum was not observed useing rMIC3-ELISA.The total coincidence rate between the ELISA and the LAT was 92.56%.[Conclusion] The ELISA is of sensitivity,short reaction time,high specificity,and reproducibility,and would be a useful tool for serodiagnosis and seroprevalence investigation of swine toxoplasmosis.
