不同激素组合对草麻黄愈伤组织诱导的影响
Effects of Different Hormone Combinations on Callus Induction of Ephedrae sinica Stapf
【摘要】 [目的]研究不同激素组合对草麻黄愈伤组织诱导的影响。[方法]以草麻黄子叶为外植体,在附加不同浓度和比例2,4-D、NAA、6-BA、KT的MS培养基中,比较不同激素组合对愈伤组织诱导的影响,并绘制出草麻黄愈伤组织的生长曲线。[结果]结果表明,最适合草麻黄愈伤组织诱导的培养基为MS+2,4-D2.0 mg/L+6-BA1.0 mg/L,诱导率最高为77.5%。草麻黄愈伤组织生长曲线呈S形,在第8天左右进入快速增长期,最佳继代时间在24 d左右。[结论]2,4-D与6-BA配合使用更有利于草麻黄愈伤组织的诱导。
【Abstract】 [Objective]The research aimed to study the effects of different hormone combinations on callus induction of Ephedrae sinica Stapf.[Method]Segments of cotyledon of E.sinica Stapf were cultured in vitro on MS medium supplemented with different plant hormones such as 2,4-D,NAA,6-BA and KT.And the effects of plant hormone on callus induction were compared,the growth curve of E.sinica Stapf callus was drawn.[Result]The results showed that supplemented with 2,4-D 2.0mg/L+6-BA 1.0mg/L,the rate of callus induction was highest,which reached 77.5%.Growth curve of callus was S type.Callus growed rapidly after 8 days.The best time of subculture was about 24 days.[Conclusion]The combination of 2,4-D and 6-BA was much more beneficial for callus induction of E.sinica Stapf.
